Antiviral mechanism of new endoperoxide compounds showing antimalarial activity
Objectives
This project investigates the antiviral mechanisms of new compounds N-89 and N-251 against the hepatitis C virus (HCV).
The study was carried out to clarify the anti-hepatitis C virus (HCV) mechanism of new anti-HCV compounds (N-89 and its derivative N-251). First, cell lines possessing an N-89- or N-251-resistant phenotype (resistant cells) were established from N-89- or N-251-sensitive HCV-RNA-replicating cells (sensitive cells). Using these sensitive and resistant cells, the action mechanism of the compounds was analyzed. It was found that both host and viral factors contribute to approximately the same degree of resistance to these compounds. Amino acid substitutions, which might be involved in the acquisition of resistance, were detected through genetic analysis of HCVs derived from the resistant cells. Furthermore, gene candidates potentially contributing to resistance acquisition were selected via cDNA microarray analysis. In addition, it was demonstrated that these compounds show antiviral effects against Japanese encephalitis virus, hepatitis B virus, and hepatitis E virus.
