Prevent malaria infection targeting erythrocyte ATP receptor signaling: Establishment of nonresistant treatment
Objectives
The objective of this study is to identify target molecules on the erythrocyte that influence malaria parasite invasion without causing resistance. This study will focus on the role of EPAC (exchange protein directly activated by cAMP) and examine the invasion of red blood cells of malaria plasmate in the presence of activators or inhibitors.
Research conducted until 2021 revealed that, unlike the initial hypothesis that “increase in cAMP concentration in host erythrocytes and associated PKA phosphorylation are essential for erythrocyte invasion of malaria parasites,” “intraerythrocyte CAMP concentration A rise in the number rather suppresses the invasion of protozoa.” In support of the latter hypothesis, Sp-cAMP, a cAMP analog that permeates the plasma membrane, inhibits erythrocyte invasion of malaria parasites, whereas Rp-cAMP, a similarly membrane-permeable but inactive cAMP analog, inhibits erythrocyte invasion of malaria parasites. It was confirmed that the invasion of protozoa was not affected at all. It was found that there is, and are proceeding with the investigation.
