Nyssorhynchus darlingi production and infection platform – Experimental model for the study of the pathogen-vector relationship (Plasmodium vivax – Ny. darlingi) in Brazil
Objectives
This project intends to:
- Create a platform for mass creation of Ny. darlingi and making it available to the scientific community,
- Carry out the genetic characterization of the colony and the evaluation of SNPs related to resistance to insecticides in a collaboration established with Prof. Suzana Campino from the London School.
- Use the artificial membrane feeding assays, previously established by our group, to evaluate the blocking effect of the treatment for malaria established by the Ministry of Health in Brazil and
- Investigate new transmission blockers using Ny’s model. darlingi and P. vivax.
Nyssorhynchus darlingi is the main vector in the Amazon region and one of the transmitters of Plasmodium spp. most efficient in the world. In the Amazon, this is considered the primary vector in almost all states, being highly competent in the parasite transmission process to the human host. Intervention methods for malaria control, targeting the invertebrate host, whether using insecticides, sporogony cycle blockers, or exoerythrocytic cycle blockers, represent a major challenge for the scientific and health community. Since the rapid emergence and spread of insecticide-resistant mosquitoes and the scarcity of transmission-blocking drugs exacerbate this scenario. In this sense, the establishment of a mass creation of anophelines in the laboratory is extremely necessary and represents a primary condition for the development of research on malaria in a multidisciplinary context. Therefore, the Entomology group (Fiocruz Rondônia) was the second laboratory in the world to establish a highly productive colony of Ny. darlingi. This is a multidisciplinary project in partnership with important centers such as: FIOCRUZ, London School, National Institute of Health, Harvard and University of São Paulo.
