Mutations of antimalarial resistance in Equatorial Guinea and Ethiopia: differences in areas with different malaria endemicity
Objectives
- To determine the level of mutations related to resistance in the genes pfdhfr, pfdhps, pfmdr1 and pfcrt of P. falciparum, as well as in the genes pvdhfr and pvdhps of P. vivax in isolates from Equatorial Guinea and Ethiopia and compare the level of mutations for each of the species in both study areas.
- Establish if the different level of endemicity and transmission of malaria in both areas influences the level of molecular resistance detected.
- Establish a georeferenced map of the distribution of the resistant genotypes of both study and species zones.
The procedure will be the same in both study areas. Blood samples will be collected on Whatman paper, from all those individuals whose rapid malaria test has given positive results. Said samples, once dried, will be stored at -20 C for later use in the Madrid laboratory. From the whatman paper, DNA extraction will be performed by chelex-saponin and then molecular processing including: Seminested Multiplex PCR for species determination, Nested PCR for each of the genes under study related to antimalarial resistance and subsequent digestion with restriction enzymes, which will allow to know the point mutations that may exist in each of the genes and that are related to resistance to sulfadoxy / pyrimethamine, chloroquine, mefloquine, artesunate, etc.
