Functional role of antibodies of P. vivax Asymptomatic individuals in the Peruvian Amazon

Objectives

The objective of this project is to test the hypothesis that IgM and IgG antibodies from Plasmodium vivax (Pv) infected asymptomatic individuals, from the Peruvian Amazon, are capable of activating the complement system in a dependent or non-dependent manner and, inhibiting the invasion process of Pf or Pv merozoites into the erythrocyte and/or reticulocyte in in vitro and/or ex vivo conditions to a greater capacity than antibodies from Pv symptomatic individuals.

Rationale and Abstract

Several studies have described the association of IgG and IgM antibodies levels in Pv infected individuals from different malaria-endemic settings to different Pv antigens in search of seromarkers for detection of Pv recent exposure, hyponozoites reservoirs or associated risk factors among others. Large-scale analysis using protein arrays and bead-based multiplex assays pointed out the antigens: Pv Merozoite Surface Protein-8 (PvMSP8) and Pv Reticulocyte Binding protein b (PvRBP2b), as potential seromarkers for detection of Pv exposed individuals from the Peruvian Amazon, these also have been associated to acquisition of clinical immunity in Pv infected individuals and therefore are candidates for the design of a Pv vaccine. However, it remains to find out if these antigens are fundamental for the process of invasion of Pv isolates from the Peruvian Amazon and if they elicited functional and protective antibodies
