Evaluation of the combination of conditional gene expression tools in Plasmodium falciparum for functional genomic studies and screening of antimalarial potentials
Objectives
The aim of this study is to confirm and evaluate the effectiveness of a gene expression system using a fusion of destabilization domains (DDD from E. coli DHFR and DD from human FKBP12) for regulating gene expression in Plasmodium falciparum.
Gene expression systems based on destabilization domains allow rapid regulation of expression of genes of interest through reversible protein degradation. The team identified a fusion between the destabilization domains DDD, based on E. coli DHFR and DD based on human FKBP12, which possibly promotes better regulation of expression of the gene of interest than the isolated domains. The researchers intend to confirm these results and apply this system in the investigation of essential genes of P. falciparum and evaluate its applicability in detecting the toxic effect of potential antimalarials.
