Functional analysis of malaria vaccine candidate antigen GAMA
Objectives
To elucidate the molecular function of GPI-anchored merozoite antigen (GAMA), a candidate antigen for the erythrocyte stage vaccine.
Specific objectives to be carried out by the project:
- Search for GAMA-interacting molecules using a human erythrocyte surface protein library.
- Production of anti-GAMA8 monoclonal antibody (mAb);
- Measurement of protein interaction and inhibitory activity against protozoan proliferation by mAb to clarify the protein interaction essential for the molecular function of GAMA.
Malaria vaccine development is an urgent issue. Based on the wheat cell-free system, we have been energetically identifying novel erythrocyte-phase vaccine candidate molecules and searching for erythrocyte receptors using human protein libraries. In 2021, the project synthesized GST-tagged GAMA molecules and a human erythrocyte protein library using a wheat cell-free system, search for erythrocyte-side interacting molecules using the alpha screen method, and discover multiple erythrocyte-side interacting molecules. bottom. Furthermore, mice were immunized with the GAMA8 protein, and the reactivity of the antiserum to the protozoan antigen was confirmed using Western blotting and IFA. In the future, the plan is to conduct biochemical analysis based on the results obtained in 2021.
